I saw a couple of questions related to this, but I need some specific pointers. I made this guy in square stitch, and I want to attach a backing to make it into a keychain. Should I glue it? Do I need a specific glue (I have hot glue)? Or should I stitch it, and if so what would be the best way to do that?

You could glue or stitch it. Leather would be a good, durable backing, and doesn't fray. If you aren't into using leather, felt would work but wouldn't be as durable. If you go the glue route, I would recommend using E6000. Glass beads are non-porous, so hot glue wouldn't adhere to them very well. E6000 is meant for non-porous (glass, metal) and porous (wood, paper, leather, fabric) materials.
